Background: The prevailing methods for the genomic DNA extraction of Sclerotium rolfsii from its mycelium mat is often time consuming and yields poor quality quantity owing to presence outrageous magnitude mucilage polysaccharides. Methods: A new fast track method isolation resting structure (sclerotia) S. by modifying CTAB deprived supplementation proteinase K has been standardized. Result: protocol produced 500ng sclerotia with purity vacillating 1.7 1.9 as confirmed A260/A280 A260/A230 spectrophotometric documentation. extracted commissioning was efficaciously used further downstream reactions/process like PCR-RAPD, PCR-ISSR ITS amplification rDNA-ITS region.
